How to Verify a Certificate of Analysis

A Certificate of Analysis (COA) is the record that ties a specific batch of a research compound to an independent lab's measurement of what it actually contains. This guide covers what a COA verifies, how to read the purity and identity results, and how to tell a verifiable report from one that isn't.

What does a Certificate of Analysis verify?

A COA verifies three things: the compound's identity, its HPLC purity percentage, and the specific batch (lot) it was tested from.

Those three facts — identity, purity, lot — are what a per-batch COA is for. A report that omits any one of them is not verifying the vial it claims to describe.

How do you read an HPLC purity trace?

One tall, narrow peak means one substance dominates the sample; that peak's area, divided by the total peak area, is the purity percentage.

High-performance liquid chromatography (HPLC) separates a sample's components by how long each takes to pass through a column, then plots each as a peak over time. A clean single peak at the expected retention time, with minimal area under any other peak, is what a high-purity result looks like on the trace itself — not just in the summary number printed beside it.

What confirms a compound's identity on a COA?

Identity is confirmed by mass spectrometry, which measures a sample's molecular weight against the compound's known reference mass.

Purity alone does not prove identity — a sample can be 99% pure and still be the wrong molecule. Mass spectrometry (MS) fragments the sample and measures the resulting ion masses; a match to the compound's published molecular weight and formula is the identity confirmation, independent of the HPLC purity result.

Third-party lab vs in-house testing — what's the difference?

A third-party lab has no financial stake in the result; an in-house test is run by the seller itself and cannot self-verify.

Independent COA-comparison platforms exist specifically because in-house results are not independently verifiable — the lab, the seller, and the interested party are the same entity. A third-party lab's accreditation and its lack of a stake in the sale are what make its result checkable against an outside standard.

What are red flags in a COA?

No lot number, no lab name or accreditation, a purity figure with no visible trace, or a report whose batch code doesn't match the vial.

A trustworthy COA is verifiable, not just stated: it names the testing lab, carries a lot number that appears on the physical vial, and shows the underlying trace or spectrum — not only a headline purity percentage. Any COA missing that chain from lab → lot → vial is a document that cannot actually be checked against what shipped.
